Summaries

Actin disruptors

We expect actin disruptors to create a loss of F-actin structures, no more filaments especially on the outside, rather more cytoplasmic fluorescence, sometimes even aggregates. We expect the cells to get rounder and have fewer protrusions, possibly even some blebbing: this could be visible in the tubulin channel even though no direct changes are expected in the microtubules.

Microtubule destabilizers

We expect microtubule destabilizers to create loss of microtubules, creating a diffuse cytoplasmic signal. We expect potentially some indirect effect on actin, with an increase of actin in stress fibers and cortical actin. We expect cells to be more compact in shape, possible rounder. We expect less spreading out of the cell, less dynamic margins, fewer processes.

Protein synthesis

We expect protein synthesis inhibitors to sometimes disorganize microtubules, and sometimes cause stress-signals in the actin signals (e.g. less dynamic structures, especially at the leading edge). We expect the cells to be less dynamic, potentially shrink and bleb or atrophy.

Protein degradation

Note

It may not be possible to summarize this one. Will have to correlate the effects that we're seeing drug by drug.

No immediate actin changes; possibly aberrant accumulations over time
